I'm thinking of picking up TL2.  Are abilities mapped to mouse buttons, hot keys, or both?

Both. You can assign 2 abilities to left/right mouse button. The abilities on your taskbar can't be activated by mouseclicking though. You need to press their hotkeys. Clicking them with mousebutton lets you change the assigned ability/consumable.
