It might be a dumb question but hey, i'll ask it anyways.

Can i upgrade a trial account/buddy key to a full account using a game card?

No.

 

You need to buy the game first (either box or purchase of a game key will work) and then you get your free 30 days of play. Then you can use game cards for your monthly subscription, actually it's 60 days on the game card here in Europe, not sure about the US.

Edit to add that you can continue with the characters you made during your trial period if you want.
